What is AOD-9604 peptide?

AOD-9604 is a synthetic peptide of 16 amino acids, based on the C-terminal region of human growth hormone (amino acids 176–191). It differs from the original fragment through an additional tyrosine at the N-terminus, which improves molecular stability. The peptide was developed by the Australian company Metabolic Pharmaceuticals in the late 1990s.

The aim was to separate growth hormone's effects on fat metabolism from its other effects. AOD-9604 is therefore not growth hormone and is not a secretagogue like GHRP-6 or ipamorelin.

AOD-9604 for weight loss: what the studies found

AOD-9604 is most commonly associated with weight loss in search queries, so here is an unvarnished account of its research history:

• Preclinical studies in mice found increased fat oxidation and less weight gain.

• Early clinical trials (phase II) produced modest results, which supported moving on to a larger trial.

• Phase IIb trial (around 2007) failed to meet its primary endpoint: weight loss was not significantly different from placebo. Development as an obesity treatment was then discontinued, and Australia's TGA rejected this indication.
